In Vivo Spatial Transcriptomics for Bleeding-free Profiling Human Internal Organs

Abstract

This preprint presents ENDO-Genome, a system that integrates a nanoarrayed biochip with standard endoscopy for minimally invasive in vivo transcript sampling from internal organs without tissue biopsy. The pressure-calibrated approach profiled 55 messenger RNA targets from multiple intestinal locations during a five-minute procedure. In a clinical study of 15 patients with Crohn's disease, 47 procedures were completed without reported complications and revealed spatially heterogeneous inflammatory and RNA-regulatory programs, supporting bleeding-free live spatial transcriptomic analysis.
