Why immune states differ

Colorectal tumours differ in their immune environments and responses to treatment. MotifCC integrates expression, mutation and methylation measurements to investigate whether these differences define reproducible disease states and suggest biologically testable interventions.

The study analysed 627 TCGA cases and 977 cases from the Chinese COCC cohort. It identified three immune subtypes with distinct molecular characteristics, microenvironments and outcomes. One subtype combined strong WNT pathway activation with an immune-cold state and poor prognosis.

From classification to experimental investigation

The WNT-related finding motivated experiments in cells, patient-derived organoids and animal models. These experiments supported WNT inhibition as a candidate strategy for improving antitumour immunity and immunotherapy response.

MotifCC connects molecular classification with a specific intervention hypothesis and experimental testing. The intervention evidence is preclinical; clinical treatment effects are a subsequent research question.
