Our team published PIANOS in Nature Communications to address a long-standing challenge in colorectal cancer prognosis: model instability across platforms and cohorts.
PIANOS is a platform-independent, normalization-free single-sample classifier built on pathway-level ranking and k-TSP logic. It enables robust patient-level risk assessment without cross-cohort normalization.
Key points:
- Validated in 15 independent cohorts from 8 countries (n=3666).
- Achieved strong overall prognostic performance and outperformed a large set of published CRC models.
- Revealed biologically distinct risk groups with different potential sensitivity to chemotherapy, anti-angiogenic therapy, and immunotherapy.
